Regulatory bodies like the FDA, EMA, and ICH (e.g., ICH Q2, ICH M10) require ELISAs used for quantitative purposes to be validated for:
- Accuracy & precision
- Sensitivity (LOD & LOQ)
- Linearity and dynamic range
- Matrix effects
- Intra-/inter-assay reproducibility
- Stability of reagents & samples

Step 1: Define the Analytical Purpose
Is the assay intended for:
- Pharmacokinetic (PK) studies of biosimilars?
- Immunogenicity detection for therapeutic antibodies?
- Small molecule quantification in human serum?
- Diagnostic biomarker detection?
Each use case demands a different validation depth. For example, a PK ELISA for Adalimumab needs precise curve fitting, whereas a vitamin D ELISA for diagnostics may prioritize cutoff sensitivity and sample throughput.
Step 2: Linearity & Calibration Curve
Each Denovo DeQuanto® ELISA undergoes curve fitting with R² ≥ 0.98, using validated standards across:
- Adalimumab (PK1002): 5.2 – 200 ng/mL
- Bevacizumab (PK1005): 0.1 – 3.1 µg/mL

This ensures accurate back-calculation across the full dynamic range, especially at clinical low-end dose levels.
Step 3: Sensitivity (LOD & LOQ)
We calculate LOD using blank sample mean + 3 SD, and confirm LOQ with CV ≤ 20%.

Step 4: Precision & Reproducibility
We evaluate:
- Intra-assay precision: %CV across replicates within the same plate
- Inter-assay precision: %CV across 3–5 days, analysts, or lots
Accepted thresholds:
- ≤15% CV for medium/high QC
- ≤20% CV for LLOQ samples

Step 5: Matrix Compatibility & Interference
Every Denovo ELISA is validated in human serum and plasma, using:

- Parallelism testing
- Cross-reactivity evaluation (especially for immunogenicity ELISAs)

Step 6: QC & Stability Testing
