Super Sensitive TMB Substrate (1X), Ready to Use for ELISA
Scientific Background
DeQuanto® Super sensitive TMB Substrate is a single-component, ready-to-use proprietary formulation containing 3,3’,5,5’-tetramethylbenzidine (TMB). It has wide application as chromogenic substrate for Peroxidase (HRP) labelled conjugates. Super Sensitive TMB substrate provides high signal and high sensitivity for ELISA. The Super Sensitive TMB Substrate produces a soluble blue colour with HRP, which can be read at 620-650 nm. This reaction can be stopped with 2N H2SO4 or 2N HCl or 1M H3PO4. The optical density of the resulting colour solution can be read at 450 nm in an ELISA reader/spectrophotometer
Direction for Use
- Reagent is clear to pale blue solution.
- Bring the super sensitive TMB solution to Room temperature before use.
- Pipette 100 µl/well Super Sensitive TMB substrate into each well of the microtiter plate.
- Incubate the plate in dark for 15-30 minutes and monitor the colour development.
- Read the blue colour at 620-650 nm in ELISA reader/spectrophotometer.
- Stop the reaction using 2N H2SO4 or 1M H3PO4.
- Read optical density at 450 nm after stopping the reaction. This yellow colour is stable for 30 minutes.
Precautions
- Do not dilute the reagent.
- Avoid direct exposure to light
